El género Pestivirus de la familia Flaviviridae incluye diferentes especies virales que causan pérdidas importantes a la industria ganadera en todo el mundo afectando bovino, ovino/caprinos y cerdos. La detección rápida, sensible y específica de estos agentes es por lo tanto esencial para el diagnóstico y control de estas infecciones virales. Se desarrolló y estandarizó un ensayo de RT-PCR para la detección específica de pestivirus en suero basado en una pareja de cebadores que amplifica un fragmento de 119 pb de una región altamente conservada en el extremo 5´UTR del genoma ARN del virus. Este ensayo mostró ser propósito específico, sensible y altamente repetible. |

O objetivo do trabalho foi efetuar um levantamento previo da ocorrencia de bovinos portadores de diversos virus causadores de doencas da reproducao no Pantanal, para, em fase posterior, avaliar o seu efeito sobre a fertilidade do rebanho. Entretanto, e de vital importancia que o produtor pantaneiro saiba mais sobre essas doencas e que ja foram diagnosticadas na regiao, para que possa ficar atento quanto ao aparecimento das mesmas em suas propriedades ou para que adote criterios quando da aquisicao de reprodutores. A two-month-old lamb showing signs of severe neurological disease characterized by muscular tremors, hypermetria, and motor incoordination was submitted to the Veterinary Pathology Laboratory - Universidade Federal do Rio Grande do Sul, Brazil. At necropsy, the major findings were a marked reduction of the size of the cerebellum and bilateral dilatation of the lateral ventricles. Microscopically, areas of cellular disorganization in the cerebellar cortex, reduction of the granular layer of cerebellum associated with decreased density of cells, and the presence of large cytoplasmic vacuoles in the molecular layer were observed. Serum neutralization tests (SN) were performed against classical swine fever virus (CSFV), bovine viral diarrhea virus (BVDV) and border disease virus (BDV) on samples of swine serum collected for screening of antibodies to CSFV, in order to determine the SN value as a differential serological test. Ninety-nine sera out of a sample of 16,664 were positive for antibodies to pestiviruses in an ELISA test which did not distinguish antibodies to different pestiviruses. When submitted to SN, 81 sera were positive for CSFV antibodies only. In 17 sera, crossreactive antibodies to either CSFV, BVDV or BDV were detected. Abstract The open reading frame of a Brazilian bovine viral diarrhea virus (BVDV) strain, IBSP4ncp, was recombined with the untranslated regions of the reference NADL strain by homologous recombination in Saccharomyces cerevisiae, resulting in chimeric full-length cDNA clones of BVDV (chi-NADL/IBSP4ncp#2 and chi-NADL/IBSP4ncp#3). The recombinant clones were successfully recovered, resulting in viable viruses, having the kinetics of replication, focus size, and morphology similar to those of the parental virus, IBSP4ncp.
